ABSTRACT

Preeclampsia is a major reason of morbidity and mortality in pregnant women and perinatal fetus. Hence, it is of prime importance that diagnostic markers are defined to predict chances of preeclampsia in pregnant women. It has been previously shown that microRNA (miRNA)-376c expression is decreased in the placenta of preeclampsia patients at term. Even though this decrease was not mimicked in the placenta at the pre-term stage, miR-376c expression was decreased in the plasma of these patients as early as the second trimester. Plasma and placenta specimens were obtained from pregnant women having unifetal gestation undergoing perinatal care between January 2014 and December 2016 (n=49). Early trimester placentas were collected from patients undergoing terminated pregnancies through dilation and curettage procedure. Our results showed that in addition to miR-376c, miR-441 levels were decreased in the placenta of preeclampsia patients, and this decrease occurred both at pre-term and at term. This decrease is also mimicked in the plasma levels at both early and late weeks of pregnancy, highlighting that miR-441 levels can serve as a diagnostic marker of risk of preeclampsia in pregnant women. Overexpression of the miR-441, as well as miR-376c, promoted cell viability, migration, and invasion in the human immortalized cytotrophoblast cell line HTR8/SVneo, indicating that their decrease in pregnant women would result in anomalous apoptosis and functional imbalance resulting in premature abortion and other complications. MiR-441 level can thus potentially serve as diagnostic marker of preeclampsia in pregnant women.

ABSTRACT

5-methylcytosine (5-mC) and 5-hydroxymethylcytosine (5-hmC) are modified cytosines found in mammals that are involved in the regulation of gene expression. The aim of this study was to characterize the global patterns of 5-mC and 5-hmC of the fetal placenta of Nellore cattle as well as blood and sperm as controls. 5-mC and 5-hmC levels were determined using MethylFlash Methylated/Hydroxymethylated DNA Quantification Kit, respectively. Placenta tissues showed lower levels of 5-mC and 5-hmC compared to sperm. The male cotyledon showed higher levels of 5-hmC than the female. For the first time, the levels of 5-mC and 5-hmC in Bos taurus indicus were characterized, which may contribute to our understanding of the mechanisms of epigenetic regulation in the placenta. The presence of 5-hmC in somatic tissues suggest that 5-hmC has its own biological function and it is not only a byproduct from the oxidation of 5-mC. These results may be of interest in ARTs, especially in cloning in the diagnosis/prognosis of aberrant placentation and the viability of pregnancies.(AU)


5-metilcitosina (5-mC) e 5-hidroximetilcitosina (5-hmC) são citosinas modificadas encontradas nos mamíferos que estão envolvidas com a regulação da expressão gênica. O objetivo do presente estudo foi caracterizar os padrões globais de 5-mC e 5-hmC em placenta fetal de animais da raça Nelore, assim como em sangue e espermatozoides, usados como controles. Os níveis de 5-mC e 5-hmC foram determinados usando os kits MethylFlash Methylated/Hydroxymethylated DNA Quantification, respectivamente. Tecidos placentários apresentaram menores níveis de 5-mC e 5-hmC quando comparados com espermatozoides. Cotilédones de machos apresentaram maiores níveis de 5-hmC do que os de fêmeas. Os níveis de 5-mC e 5-hmC em animais Bos taurus indicus foram caracterizados pela primeira vez, o que pode contribuir para o nosso conhecimento sobre a regulação dos mecanimos epigenéticos na placenta. A presença de 5-hmC em tecidos somáticos sugerem que essa base pode ter sua própria função biológica, sendo não somente um sub-produto da oxidação da 5-mC. Esses resultados podem ser de interesse nas Tecnologias de Reprodução Assistida, especialmente na clonagem, no diagnóstico/prognóstico de placentação aberrante e viabilidade da progênie.(AU)

ABSTRACT

Newborn's health is directly related to gestational conditions and placental efficiency. The aims of this study were: (1) To evaluate hematological and biochemical parameters of foals born from mares with placentitis at birth and at 24h of age, (2) to verify if placental pathology had any influence on neonatal maturity degree through hematological and biochemical response of those foals. According to placental findings (control and placentitis) and neonatal maturity degree (mature and immature), foals were divided into three groups: (1) Control group (n=22), foals born from mares with placentitis and classified as (2) Mature (n=26), and (3) Immature (n=10). The hematocrit and plasma concentration of fibrinogen, total plasma protein, white blood cells count, lactate, glucose, creatinine, urea, albumin, bilirubin, triglyceride, cholesterol, calcium, phosphorus, magnesium, aspartate aminotransferase (AST), creatine kinase (CK), alkaline phosphatase (ALP), and gamma-glutamyltransferase (GGT) were measured. Placental features were significantly different between neonatal maturity degree (P=0.001). Mares that had acute placentitis foaled more immature neonates (n=8/10; 80%). Concentrations of fibrinogen (P=0.003), creatinine (P=0.021), total cholesterol (P=0.014), AST (P=0.001), GGT (P=0.002), total (P=0.001) and unconjugated bilirubin (P=0.010) were higher at birth in the Immature group, whereas albumin levels were lower (P=0.002). Foals born from mares with placentitis presented hyperlactatemia at 24h of age (P=0.002). Acute placentitis had an influence on the neonatal maturity, allowing an accelerated but incomplete fetal maturation. The monitoring of lactate, fibrinogen, creatinine, bilirubin, cholesterol, albumin, AST, and GGT levels, associated with clinical, physical, and behavior evaluation may contribute as indicators of neonatal maturity.(AU)


A saúde do neonato está diretamente relacionada às condições gestacionais e eficiência placentária. Os objetivos deste estudo foram: (1) avaliar parâmetros hematológicos e bioquímicos de potros nascidos de éguas com placentite ao nascimento e com 24h de vida e (2) verificar se a patologia placentária exerceu influência no grau de maturidade através da resposta hemato-bioquímica destes neonatos. De acordo com os resultados histopatológicos placentários (controle e placentite) e grau de maturidade neonatal (maturo e imaturo), os potros foram divididos em três grupos: grupo controle (n=22); e potros nascidos de éguas com placentite classificados como (2) maturos (n=26) e (3) imaturos (n=10). Foi avaliado hematócrito e concentrações sanguíneas de fibrinogênio, proteína plasmática total, leucócitos totais, lactato, glicose, creatinina, uréia, albumina, bilirrubinas, triglicerídeos, colesterol, cálcio, fósforo, magnésio, aspartato aminotransferase (AST), creatina quinase (CK), fosfatase alcalina (FA) e gama glutamiltranferase (GGT). As características placentárias foram significativamente diferentes entre os graus de maturidade neonatal (P=0.001). Éguas com placentite aguda produziram mais potros imaturos (n=8/10; 80%). No nascimento, os potros imaturos apresentaram maiores concentrações de fibrinogênio (P=0,003), creatinina (P=0,021), colesterol total (P=0,0014), AST (P=0,001), GGT (P=0,002), bilirrubina indireta (P=0,010) e total (P=0,001) e menor concentração de albumina (P=0,002). Os potros nascidos de éguas com placentite apresentaram hiperlactatemia com 24h de vida (P=0,002). A placentite aguda exerceu influência na maturidade neonatal, permitindo uma maturação fetal acelerada, porém, incompleta. Mensurações dos níveis sanguíneos de lactato, fibrinogênio, creatinina, colesterol total, AST, GGT, bilirrubinas e albumina, associado à avaliação clínica, física e comportamental, podem contribuir como indicadores de maturidade neonatal.(AU)

ABSTRACT

The sequestration of infected erythrocytes in the placenta can activate the syncytiotrophoblast to release cytokines that affect the micro-environment and influence the delivery of nutrients and oxygen to fetus. The high level of IL-10 has been reported in the intervillous space and could prevent the pathological effects. There is still no data of Th17 involvement in the pathogenesis of placental malaria. This study was conducted to reveal the influence of placental IL-17 and IL-10 levels on fetal weights in malaria placenta. Seventeen pregnant BALB/C mice were divided into control (8 pregnant mice) and treatment group (9 pregnant mice infected by Plasmodium berghei). Placental specimens stained with hematoxylin and eosin were examined to determine the level of cytoadherence by counting the infected erythrocytes in the intervillous space of placenta. Levels of IL-17 and IL-10 in the placenta were measured using ELISA. All fetuses were weighed by analytical balance. Statistical analysis using Structural Equation Modeling showed that cytoadherence caused an increased level of placental IL-17 and a decreased level of placental IL-10. Cytoadherence also caused low fetal weight. The increased level of placental IL-17 caused low fetal weight, and interestingly low fetal weight was caused by a decrease of placental IL-10. It can be concluded that low fetal weight in placental malaria is directly caused by sequestration of the parasites and indirectly by the local imbalance of IL-17 and IL-10 levels.

ABSTRACT

Objetivos. Determinar los niveles de plomo (Pb) en gestantes y neonatos en la ciudad de La Oroya, Perú. Materiales y métodos. Se realizó un estudio transversal. La población estuvo conformada por gestantes residentes durante al menos dos años en la ciudad de La Oroya, cuando operaban normalmente las fundiciones de Pb, cobre y zinc. Se obtuvo muestras de sangre de la gestante antes del parto y luego del parto se tomaron muestras de placenta y sangre del cordón umbilical. El nivel de Pb de dichas muestras fue evaluado por espectrometría de absorción atómica con horno de grafito. Se determinaron regresiones cuadráticas y correlaciones de Pearson. Resultados. Se evaluó cuarenta partos normales. Los niveles promedio de Pb en la sangre materna, cordon umbilical y placenta fueron 27,4 ± 15,6 ug/dL; 19,0 ± 12,6 ug/dL y 319,0 ± 215,9 ug/100 g, respectivamente. El 67,5% de los neonatos tuvieron más de 10 ug/dL de Pb. El nivel de Pb de la sangre del cordón umbilical representó el 69,4% del nivel de Pb en sangre materna. Los coeficientes de correlación entre el Pb de la sangre materna y cordón umbilical, sangre materna y placenta, placenta y sangre del cordón umbilical fueron 0,36; 0,48 y 0,33, respectivamente. Conclusiones. Cuando funcionaba la fundición, las gestantes y recién nacidos tenían concentraciones elevadas de Pb en sangre. Las concentraciones de Pb en la placenta y cordón umbilical tuvieron una correlación moderada con el Pb de la sangre materna. El Pb de la sangre materna y cordón umbilical fue 1,5 veces más alto que los de zonas más alejadas.


Objectives. To determine blood-lead (Pb) levels in pregnant women and neonates in a metallurgical city of Peru. Materials and methods. A cross-sectional study was carried out, the population of which consisted of pregnant women who had lived for at least two years in the city of La Oroya, at the time when the lead, copper and zinc foundries operated normally. A blood sample was obtained from the pregnant women before delivery and, after delivery, from the neonate’s umbilical cord and placenta. Lead levels in those samples were measured by graphite furnace atomic absorption spectrophotometry. Pearson’s squared regressions and correlations were determined. Results. Forty normal deliveries were studied. The average blood lead levels in the mother, umbilical cord and placenta were 27.4 ± 15.6 ug/dL; 19.0 ± 12.6 ug/dL and 319.0 ± 215.9 ug/100 g, respectively. 67.5% of the neonates registered levels higher than 10 ug/dL. Pb levels in the umbilical cord accounted for 69.4% of Pb level in maternal blood. The correlation coefficients between Pb levels in maternal blood and umbilical cord, maternal blood and placenta, and placenta and umbilical cord were 0.36; 0.48 and 0.33, respectively. Conclusions. At the time the foundry operated, pregnant women and newborns had high blood lead concentrations. Pb concentrations in the placenta and umbilical cord had moderate correlation with the Pb levels in maternal blood. Pb levels in maternal blood and umbilical cord were 1.5 times as high as in more distant areas.

ABSTRACT

Interleukin-10 (IL-10) appears to be the key cytokine for the maintenance of pregnancy and inhibits the secretion of inflammatory cytokines such as tumor necrosis factor-α (TNF-α). However, there are no studies evaluating the profile of these cytokines in diabetic rat models. Thus, our aim was to analyze IL-10 and TNF-α immunostaining in placental tissue and their respective concentrations in maternal plasma during pregnancy in diabetic rats in order to determine whether these cytokines can be used as predictors of alterations in the embryo-fetal organism and in placental development. These parameters were evaluated in non-diabetic (control; N = 15) and Wistar rats with streptozotocin (STZ)-induced diabetes (N = 15). At term, the dams (100 days of life) were killed under anesthesia and plasma and placental samples were collected for IL-10 and TNF-α determinations by ELISA and immunohistochemistry, respectively. The reproductive performance was analyzed. Plasma IL-10 concentrations were reduced in STZ rats compared to controls (7.6 ± 4.5 vs 20.9 ± 8.1 pg/mL). The placental scores of immunostaining intensity did not differ between groups (P > 0.05). Prevalence analysis showed that the IL-10 expression followed TNF-α expression, showing a balance between them. STZ rats also presented impaired reproductive performance and reduced plasma IL-10 levels related to damage during early embryonic development. However, the increased placental IL-10 as a compensatory mechanism for the deficit of maternal regulation permitted embryo development. Therefore, the data suggest that IL-10 can be used as a predictor of changes in the embryo-fetal organism and in placental development in pregnant diabetic rats.

AIM: To investigate the relationships, if any, between elemental content of the placenta with age of mother, birthweight and the Apgar scores of a neonate. METHODS: Placental samples were collected, stored at -20ºC and then dried and analysed using neutron activation with the SLOWPOKE II reactor at the International Centre for Environmental and Nuclear Sciences (ICENS). A questionnaire was administered at the time of delivery to determine the level of fish consumption, numbers of dental amalgam fillings and use of cough syrups. Placental concentrations of bromine, calcium, chlorine, iron, mercury, potassium, rubidium, selenium, sodium and zinc were determined. RESULTS: The placentae of 52 Jamaican mothers with a mean age 29 years (range 18-42 years) delivering singleton neonates with a mean birthweight of 3.1 kg (1.3 - 5.5 kg) at term were collected. With the exception of iron and bromine, all results for elemental determinations are very similar to those found elsewhere. Correlation was observed for Apgar 2 (5 minutes), calcium and zinc with birthweight, with p-values of 0.002, 0.007 and 0.07, respectively. Negative correlation was observed for the Apgar 2 and potassium (p = 0.056) and age of mother at birth and bromine (p = 0.02). The mercury concentration in the measured placentae (7.29 ±9.1 µg/kg w/w) was slightly lower than the mean concentration found in the literature (8 µg/kg w/w). Approximately 93% of the measured placentae in this study are below the derived placentae upper limit of 22 µg/kg. Of the 7% above the upper limit none exceeded the conservative estimated limit of 115 /µg/kg at which neural developmental problems start. CONCLUSION: The significant associations noted are of unknown clinical relevance and need further study.


OBJETIVO: Investigar las relaciones que pudieran existir entre el contenido elemental de la placenta y la edad de la madre, el peso al nacer, y la puntuación Apgar del neonato. MÉTODOS: Muestras de placenta fueron recogidas, almacenadas a -20ºC y entonces secadas y analizadas usando la activación neutrónica con el reactor SLOWPOKE II del Centro Internacional de Ciencias Medioambientales y Nucleares (ICENS). A la hora del parto se aplicó una encuesta para determinar el nivel de consumo de pescado, el número de amalgamas dentales y el uso de jarabes para la tos. Se determinaron las concentraciones placentales de bromo, calcio, cloro, hierro, mercurio, potasio, rubidio, selenio, sodio y cinc. RESULTADOS: Se recogieron las placentas de 52 madres jamaicanas con una edad promedio de 29 años (rango 18-42 años) que parieron neonatos únicos con un peso promedio de 3.1 kg (1.3-5.5 kg) a término. Con la excepción del hierro y el bromo, todos los resultados para las determinaciones elementales son muy similares a los hallados en otras partes. Se observó una correlación de Apgar 2 (5 minutos), el calcio y el cinc con el peso al nacer, los valores p de 0.002, 0.007 y 0.07 respectivamente. Se observó una correlación negativa de Apgar 2 y el potasio (p = 0.056) y la edad de la madre a la hora del alumbramiento, con el bromo (p = 0.02). La concentración de mercurio en las placentas medidas (7.29 ± 9.1 w/w de µg/kg) fue ligeramente más baja que la concentración promedio hallada en la literatura (8 µg/kg w/w). Aproximadamente 93% de las placentas evaluadas en este estudio están por debajo del límite superior de 22 µg/kg. Del 7% por encima del límite superior, ninguna excedió el límite conservador estimado de 115 µg/kg en el cual comienzan los problemas del desarrollo neural. CONCLUSIÓN: Se desconoce la importancia clínica de las asociaciones significativas observadas y se requiere más estudio.

ABSTRACT

Estudou-se a distribuição espaço-temporal do fator de crescimento fibroblástico básico (bFGF), do receptor 1 do fator de crescimento fibroblástico (FGFR1) e do receptor 2 do fator de crescimento fibroblástico (FGFR2) na placenta bubalina, correlacionando-a à proliferação celular. Para a detecção do bFGF, FGFR1, FGFR2 e antígeno Ki-67, colheram-se 12 placentas de búfalas nos terços inicial, médio e final da gestação, em abatedouros, e realizaram-se testes de imunoistoquímica. Detectou-se e avaliou-se a expressão do bFGF, do FGFR1, do FGFR2 e do antígeno Ki-67 ao longo da gestação. No compartimento fetal da placenta, observaram-se correlações positivas entre a expressão do bFGF e Ki-67, entre FGFR1 e Ki-67 e entre FGFR2 com Ki-67 (r=0,313, 0,358 e 0,384, respectivamente). No epitélio e estroma maternos observaram-se altas correlações entre FGFR1 e Ki-67 (r=0,739 e r=0,511, respectivamente). Os resultados sugerem envolvimento do bFGF, FGFR1 e FGFR2 na proliferação do trofoblasto enquanto no compartimento materno da placenta bubalina apenas o FGFR1 atuaria como modulador dessa atividade.


The space-temporal expression of basic fibroblast growth factor (bFGF), fibroblast growth factor receptor 1 (FGFR1) and fibroblast growth factor receptor 2 (FGFR2) in buffalo placenta and correlation to proliferative activity was studied. For the localization of bFGF, FGFR1, FGFR2 and Ki-67, 12 buffalo placentas from initial, middle and final gestational thirds were collected and immunohistochemistry tests were performed. Expression of bFGF and its receptors was detected and analyzed from the initial third until the end of gestation. In the fetal compartment, positive correlations were observed between the expression of bFGF and Ki-67, FGFR1 and Ki-67, besides FGFR2 and Ki-67 (r=0.313, 0.358 and 0.384, respectively). High correlations were found between FGFR1 and Ki-67 in maternal epithelium and stroma (r=0.789 and r=0.511, respectively). The results suggest that bFGF, FGFR1 and FGFR2 may be involved in the modulation of trophoblast proliferation, whereas maternal compartment proliferation in the buffalo placenta would only be modulated by FGFR1.

ABSTRACT

To investigate the effect of placental isoferritin (PLF) on mouse embryo development in vitro, mice 2-cell embryos were co-cultured with human first trimester decidual cells at different concentrations of PLF in vitro. The following changes of the above system were observed under an invert microscope and the number of embryos were recorded and the embryos were classified. The results showed there was no significant difference in the percentage of embryos development to 4-cell, 8-cell and morula (P>0.05). PLF at the doses of 10 and 100 U/mL significantly enhanced more embryos development to the blastocyst and hatching blastocyst (P0.05). It was concluded that PLF at the concentration of 10-100 U/mL had no significant effects on the early development of mice embryos, however, PLF could promote the growth, differentiation, and hatching of preimplantation blastocysts.

ABSTRACT

This cross-sectional study was to assess the nutrients in terms of protein, fat, minerals, and hormones in heat-dried human placenta. Thirty heat-dried human placentas, 15 from male and 15 from female, were analyzed for protein (amino acids), fiber, fat, moisture, minerals (sodium, potassium, phosphorus, calcium, iron, magnesium, zinc, copper, manganese), hormones (estradiol, progesterone, testosterone, growth hormone). Heat-dried female human placentas had slightly higher fiber content than male, but protein and fat components were not different. Mineral levels in placentas were high especially sodium, potassium and phosphorus. There were no significant differences in the amount of minerals and hormonal profile between female and male placentas. However, hormone levels in heat-dried placenta were low compared to physiologic level in human beings. The results of this study suggest that the amount of nutrients particularly protein and minerals in heat-dried human placentas were enriched.

El nacimiento prematuro del neonato representa más de 80 por ciento de muertes perinatales, no atribuibles a malformaciones congénitas. A la fecha se desconocen las causas que originan el desencadenamiento de trabajo de parto pretérmino como a término. Evidencias experimentales involucran diversas citocinas inflamatorias en el inicio y/o mantenimiento del evento. El objetivo del trabajo fue determinar la concentración de TNF-alfa e IL-1beta en los compartimentos vasculares retroplacentario, fetal y materno en mujeres con embarazos a término y pretérmino que presenten trabajo de parto, y en mujeres con embarazos a término sin trabajo de parto. La determinación de las citocinas TNF-alfa e IL-1beta, se llevó a cabo mediante ensayos inmunoenzimáticos comerciales. Los resultados obtenidos de las citocinas aquí estudiadas, no mostraron diferencia estadística alguna (prueba t) al comparar los diferentes compartimentos intra e intergrupo. Sin embargo, la concentración de TNF-alfa muestra una tendencia a encontrarse en mayor proporción el ambiente fetal y retroplacentario, en los grupos con trabajo de parto (a término y pretérmino), comparada con el valor del grupo a término sin trabajo de parto. Del mismo modo, la concentración de IL-1beta mostró una tendencia de mayor concentración en los grupos de mujeres con trabajo de parto (a término y pretérmino), pero sólo en el compartimento retroplacentario. Las tendencias encontradas nos permiten proponer al compartimento retroplacentario y fetal, como los sitios blanco donde se sintetiza TNF-alfa e IL-1beta. Asimismo, la observación de la compartamentalización de ambas citocinas, parece indicar la existencia de un gradiente de síntesis que identifica al feto como productor inicial de TNF-alfa.

ABSTRACT

Intrauterine growth retard (IUGR) continues to be a significant perinatology problem at the end of this century. The nature of the etiologic agent, the time when the attack occurred during pregnancy and its duration affect the type of IUGR. Objective: To study the evolution of fetal pancreas and placenta between the 18th and 2 1st day of pregnancy in rats submitted to maternal protein-calorie restriction. Design: Randomized controlled trial on laboratory animal. Sample: Forty-one normoglycemic pregnant Wistar rats. Intervention: Rats were divided into six experimental groups according to their access to food and date of cesarean section (18th or 21 st day): control with free access to food; diet restricted to 25 per cent introduced on 1 st day of pregnancy; and diet restricted to 25 per cent after the 3rd day of pregnancy. Main measurements: Newborn weight, placenta weight, histopathological study (morphological histochemistry). Results: Maternal protein-calorie malnutrition caused intrauterine growth retard (IUGR) after the 18 th day of pregnancy. Dietary restriction did not interfere with the morphology of the fetal pancreas and theimmunohistochemical study of the placenta showed that glycogen stores were decreased between the 18 th and 21 st day in the control group and in a diet restricted to 25 per cent from the first day of pregnancy. Dietary restriction after the 3rd day of pregnancy led to low placental glycogen concentrations on the 18 th day and disappearance on the 21 st day. Conclusion: The pathophysiology of IUGR due to maternal protein-calorie restriction in rats is related to lower placental weight and low placental glycogen stores.

ABSTRACT

A relatively simple and rapid method is described for the isolation of basal cell membranes (BCM) from the human placenta at term which showed considerable improvement in the yield, purity and membrane characteristics as compared to the earlier described methods. The method is based on thorough washings of the syncytium in balanced salt solution, selective grinding, hypotonic lysis, sonication, incubation with EDTA and then more conventional differential centrifugation and ultracentrifugation. The isolated material showed smooth surfaced vesicular structure of various sizes as revealed by both positive and negative staining and transmission electron microscopic analysis. The membrane was highly enriched in Na+/K+, Ca2+ and Mg2+ dependent ATPase activities while the cross contamination with brush border surfaces was low as revealed by the marker enzyme assays specific for the brush border membrane (BBM) such as the disaccharide hydrolases, aminopeptidase and alkaline phosphatase. The membrane showed a relatively low lipid/protein ratio and the lipid composition represented by a variety of phospholipids (phosphatidyl choline, phosphatidyl ethanolamine, sphingomyelin, phosphatidyl inositol and phosphatidyl serine), neutral lipids (cholesterol, triacyl glycerol, free fatty acids) and glycosphingolipids (ganglioside, cerebroside and sulfatide). It also contained plasmalogens. On SDS-PAGE analysis and Coomassie blue staining reaction, the isolated membrane showed 14 major bands with as many minor ones with a molecular weight ranging between 30-110 kDa.

ABSTRACT

The lipid composition of human placenta phospholipids, coming from 9 undernourished women that gave birth to low weight newborns and 9 well norished women, was analyzed using gas-liquid chromatography. Phospholipids of placentas coming from undernourished women, when compared to well nourished women, had significantly lower amounts of w-6 and w-3 fatty acids (40.1 ñ 1.5 vs 42.4 ñ 1.4 and 6.0 ñ 0.7 vs 7.1 ñ 1,3 per cent respectively). The calculated mean melting point was higher in placentas coming from undernourished women. In these women, the low content of polyunsaturated fatty acids and its replacement by short chain fatty acids was not able to balance the high mean melting points. The relative deficiency of essencial fatty acids, the low saturation index and the high mean melting point of undernourished women's placental phospholipids, may suggest a lower membrane fluidity and a subnormal essencial fatty acid content of fetal organs, that are essencial for normal growth and development

ABSTRACT

The usefulness as a pregnancy marker, of immunoreactivity with S100 protein antiserum in endometrial curettings and fallopian tubes during pregnancy was assessed. Twenty six placental tissues of various gestational age, two hydatidiform moles and four fallopian tubes removed for ectopic pregnancy were stained with S100 protein polyclonal antiserum by immunoperoxidase technique. Strong immunostaining was found in glands within the decidua and epithelial cells of fallopian tubes during early pregnancy. However, no S100 protein could be demonstrated in the endometrium in various phases, endometrial carcinomas, decidual glands beyond 12 wk of gestation and normal fallopian tubes. The results indicate a relationship between S100 protein and early pregnancy. Positive immunoreactivity with S100 protein antiserum in the decidual glands and fallopian tube epithelium may help in confirmation of doubtful cases of pregnancy.

Em 146 grávidas, com idade gestacional entre 28 e 41 semanas, observou-se uma concentraçäo média de zinco sérico no momento do parto de 0,713 ñ 0,406 *g/ml; a concentraçäo média do zinco nas 152 placentas analisadas foi 15,199 ñ 2,164 *g/mg e os níveis do zinco sérico do cordäo umbilical de 149 recém-nascidos foi de 1,090 ñ 0,307 *g/ml. O uso de ferro durante a gestaçäo näo demonstrou diferença significativa com relaçäo ao zinco sérico materno. A classe sócio-económica e estado nutricional materno näo alteraram significativamente os níveis de zinco materno na placenta e no recém-nascido analisados separadamente ou associando-se os dois indicadores
